The Kapa Taq DNA polymerase enzyme is based on the single-subunit, wild-type Taq DNA polymerase of the thermophilic bacterium Thermus aquaticus. Kapa Taq and Kapa Taq HotStart DNA polymerase enzymes have 5′-3′ polymerase and 5′-3′ exonuclease activities, but no 3′ to 5′ exonuclease (proofreading) activity. The enzyme has an error rate of approximately 1 error per 2.2 x 105 nucleotides incorporated.

Figure 1. A 500 bp fragment of the CCR5 gene was amplified using 100 ng, 10 ng, 1 ng or 100 pg of human genomic DNA as template. Kapa Taq HotStart enzyme exhibits improved sensitivity, specificity, and yield when compared with competitive HotStart products. All reactions were performed using the manufacturer-recommended protocols.
Figure 2. A 270 bp amplicon was amplified from mycoplasma DNA with Kapa Taq or Kapa Taq HotStart polymerase. Sensitivity was tested using a 10X template dilution series starting with 1 ng of DNA. Higher sensitivity and less primer dimers are observed when using Kapa Taq HotStart polymerase.

Kits can be stored for up to 18 months at -20˚C.
Kits include KAPA Taq (or KAPA Taq HotStart) DNA Polymerase (5 U/µL), KAPA Taq Buffer, and MgCl2 (25 mM). Kits are available with and without dNTP Mix (10 mM each). The following reaction buffers are available:
KAPA Taq Buffer A and B (10X, with 1.5 mM MgCl2 at 1X), supplied with KAPA Taq PCR Kits only
KAPA Taq Buffer with dye (10X, with 1.5 mM MgCl2 at 1X), supplied with KAPA Taq PCR Kits with dye only
KAPA Taq HotStart Buffer (5X, Mg-free), supplied with KAPA Taq HotStart PCR Kits only
KAPA Taq HotStart DNA Polymerase is also sold separately.
ReadyMix formulations (2X) of KAPA Taq DNA Polymerase are also available. These contain DNA Polymerase, reaction buffer (with and without dye), dNTPs and MgCl2 (at a final concentration of 1.5 mM).
